[Encoded by Human ANGPT2 Gene, Angiopoietin 2 is a 496-amino acid 57 kD (precursor) homodimeric secreted protein that contains a fibrinogen-like C-terminus. ANG2 is nearly 60% identical to ANG1. A 61 kD homodimeric alternative ANG2(443) product lacks 53 amino acids found in ANG2 encoded by exon B and part of the N-terminal coiled-coil domain. Like ANG2 and ANG1, secreted ANG2(443) binds endothelial TIE2 receptor tyrosine kinase, but not TIE1. ANG2(443) partially inhibits ANG2 and ANG1 binding to TIE2. As an ANG1 and TIE2 antagonist, ANG2 may counteract blood vessel maturation or stability by loosening cell-matrix contacts and inducing endothelial apoptosis.
