SHP heterodimerizes with nuclear hormone receptors ESR, RAR, RXR, TR, and NR1I3, specifically inhibiting their ligand-dependent transcriptional activation pathways. SHP may repress nuclear receptor-mediated transactivation via two steps: competition with coactivators and direct transcriptional repression.
