[HMGB1 is a very abundant and conserved 25-kD protein which plays a central role in the stabilization and/or assembly of several nucleoprotein multifunctional complexes through protein-protein interactions. HMGB1 is essential in a variety of DNA-related biological processes, e.g., V(D)J recombination, the initiation of transcription, and DNA repair. HMGB1 binds DNA without sequence specificity, but has a high affinity for bent or distorted DNA, and bend linear DNA. HMGB1 interacts with many apparently unrelated proteins by recognizing short amino acid sequences. HMGB1 is structured into two DNA-binding HMG-box domains (A and B) plus a long highly acidic C-terminal domain. The individual A and B boxes exhibit many of the structure-specific properties of the whole protein and the acidic tail modulates the affinity of the tandem HMG boxes in HMGB1 for a variety of DNA targets.
