[A gamma-lactam related, naturally occurring salinosporamide, isolated from the marine bacterium Salinospora sp. (old taxon actinomycete), with antineoplastic activity. Salinosporamide A irreversibly binds to and inhibits the 20S catalytic core subunit of proteasome, which is an ATP-dependent multi-subunit proteolytic complex that mediates the degradation of ubiquitinated proteins during normal cell growth and stress responses in order to balance cellular metabolism. Inhibition of ubiquitin-proteasome mediated proteolysis results in an accumulation of poly-ubiquitinated proteins. This leads to a disruption of cellular processes and results in cell cycle arrest, induction of apoptosis, and inhibition of tumor growth and angiogenesis. ( NCI )]
