- [Expressed by G1 cyclin-dependent kinase activated human ORC1L Gene (ORC1 Family), 861-aa 97-kDa nuclear Origin Recognition Complex Subunit 1 is the largest of 6 conserved subunits of the ORC complex that binds to the consensus ACS of Origins in an ATP-dependent manner, serves as a platform for assembly of additional initiation factors such as CDC6 and MCM proteins, initiates DNA replication, and is essential for viability. Bound to origins throughout the cell cycle, ORC and associated factors modulate local chromatin structure to facilitate replication initiation. Selectively phosphorylated during mitosis and unlike other ORC subunits, ORC1L levels vary during the cell cycle, regulated by ubiquitin-mediated proteolysis after replication initiation.
